Hypromellose (INN), short for hydroxypr opyl methylcel lulose (HPMC), is a semisynth etic, inert, viscoelas tic polymer used in eye drops, as well as an excipient and controlle d-delivery component in oral medicamen ts, found in a variety of commercia l products. As a food additive, Hypromell ose is an emulsifie r, thickenin g and suspendin g agent, and an alternati ve to animal gelatin. Its Codex Alimentar ius code (E number) is E464.
Chemistry of Hypromell ose
Hypromellose is a solid, and is a slightly off-white to beige powder in appearanc e and may be formed into granules. The compound forms colloids when dissolved in water. This non-toxic ingredien t is combustib le and can react vigorousl y with oxidizing agents. Hypromell ose in an aqueous solution, like methylcel lulose, exhibits a thermal gelation property. That is, when the solution heats up to a critical temperatu re, the solution congeals into a non-flowable but semi-flexible mass. Typically, this critical (congealin g) temperatu re is inversely related to both the solution concentra tion of HPMC and the concentra tion of the methoxy group within the HPMC molecule (which in turn depends on both the degree of substitut ion of the methoxy group and the molar substitut ion). That is, the higher the concentra tion of the methoxy group, the lower the critical temperatu re. The inflexibi lity, viscosity of the resulting mass, however, is directly related to the concentra tion of the methoxy group (the higher the concentra tion is, the more viscous or less flexible the resulting mass is)

When applied, a Hypromell ose solution acts to swell and absorb water, thereby expanding the thickness of the tear-film. Hypromell ose augmentat ion therefore results in extended lubricant time presence on the cornea, which theoretic ally results in decreased eye irritatio n, especiall y in dry climates, home, or work environme nts. On a molecular level, this polymer contains beta-linked D-glucose units that remain metabolic ally intact for days to weeks. On a manufactu ring note, since Hypromell ose is a vegetaria n substitut e for gelatin, it is slightly more expensive to produce due to semisynth etic manufactu ring processes . Aside from its widesprea d commercia l and retail availabil ity over the counter in a variety of products, Hypromell ose 2% solution has been documente d to be used during surgery to aid in corneal protectio n and during orbital surgery.
